Structural Control of Metabolic Flux
Organisms have to continuously adapt to changing environmental conditions or undergo developmental transitions. To meet the accompanying change in metabolic demands, the molecular mechanisms of adaptation involve concerted interactions which ultimately induce a modification of the metabolic state, which is characterized by reaction fluxes and metabolite concentrations. These state transitions a...
متن کاملGene-altered mice and metabolic flux control.
The regulation of metabolic flux is essential for life, yet our understanding of it remains incomplete. The ability to determine the role of specific enzymes in regulating metabolic flux rapidly becomes a nearly overwhelming challenge due to the various interacting pathways, multiple enzyme isoforms, and many different substrates that are involved. This complexity is further compounded by the f...
متن کاملStructural analysis of metabolic networks based on flux centrality.
Metabolic reactions are fundamental to living organisms, and a large number of reactions simultaneously occur at a given time in living cells transforming diverse metabolites into each other. There has been an ongoing debate on how to classify metabolites with respect to their importance for metabolic performance, usually based on the analysis of topological properties of genome scale metabolic...
متن کاملPhysiological control of metabolic flux: the requirement for multisite modulation.
Biochemists have long assumed that the flux through a metabolic pathway can be controlled by the activity of a key regulatory enzyme near the beginning of the pathway. We present the accumulating evidence that every step in this assumption is flawed. Instead, effective physiological control of metabolism is shown to involve simultaneous multisite modulation through action on a number of enzymes.
متن کاملFlux-P: Automating Metabolic Flux Analysis
Quantitative knowledge of intracellular fluxes in metabolic networks is invaluable for inferring metabolic system behavior and the design principles of biological systems. However, intracellular reaction rates can not often be calculated directly but have to be estimated; for instance, via 13C-based metabolic flux analysis, a model-based interpretation of stable carbon isotope patterns in inter...
